Dear Basab,
The general dictum in jyotish is that the waxing moon (cautious purists wait a few days after amavasya to begin giving the growing moon its brownie points!) and well-associated mercury are benefics. So, if mercury and saturn are placed in the same sign, that mercury may not be considered as a pure benefic. Likewise all debilitated planets have malefic properties.
Since moon is an orientation point in amala yoga, it may have been dropped out of being a benefic (when considering Lagna Amala) for conflict of interest!
But seriously, I would not take that too literally and include the moon benefic if it is waxing.
Now how to test or prove that? Well one will have to find several charts with amala yoga formed by 'benefic' waxing moon in 10th from lagna. Then one will have to rule out other benefic factors that may 'explain' the 'good' things in those charts that could get attributed to "amala". If not benefic outcome remains, then one would be confident that moon should not be treated as a benefic in the Lagna-Amala combination.
